RIPK2 (RIP2; RICK) is a death domain-containing protein kinase encoding a predicted 540-amino acid protein that contains an N-terminal serine/threonine kinase catalytic domain and a C-terminal caspase activation and recruitment domain.

  • Easily Screen and Profile RIPK2 Kinase Inhibitors
  • Includes kinase, substrate and reaction buffer
  • Use with ADP-Glo™ Assay for bioluminescent detection of kinase activity

Recombinant human RIPK2 (1-299) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. RIPK2 (RIP2; RICK) is a death domain-containing protein kinase encoding a predicted 540-amino acid protein that contains an N-terminal serine/threonine kinase catalytic domain and a C-terminal caspase activation and recruitment domain. RIPK2 is thought to regulate apoptosis induced by the FAS receptor pathway (1). RIPK2 has been shown to specifically interact with the CARD of ICE/caspase-1, and this interaction correlates with the processing of pro-caspase-1 and the formation of the active caspase-1 p20 (2). ADP-Glo Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is a substrate in a reaction catalyzed by Ultra-Glo Luciferase that produces light. The luminescent signal positively correlates with ADP amount and kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP. Kinase Enzyme System contains: Kinase: RIPK2, 10ug (Human, recombinant; amino acids 1-299). MW: ~59kDa. Substrate: Native Swine Myelin Basic Protein (MBP). Other: Reaction Buffer, DTT.
